INHIBITION OF WNT3A DIMINISHED ANGIOGENIC DIFFERENTIATION CAPACITY OF RAT CARDIAC PROGENITOR CELLS

Authors

Abstract:

Background & Aims: Myocardial infarction is a leading cause of human mortality in industrialized and developing societies. Limited restorative ability of of cardiomyocytes after ischemic changes can causes extensive damage lead to prominent chronic heart failure. At present, the application of certain drugs is touted as one of the main available approaches to inhibit the spread of the lesion and to maintain the integrity of the myocardial tissue after infarction. Today, the transplantation of stem cells to restore structure and maintain heart function has opened new hopes for clinicians in human medicine. These cells accelerate the healing process by secreting a variety of factors and differentiation into varient cell lines, including vascular cells. Here, we investigated the inhibitory role of Wnt3a factor on the process of differentiation of rat cardiomyoblast (H9C2) to endothelial cells. Materials & Methods: In this experimental study, rat cardiomyoblast (H9C2) were expanded in DMEM/HG and exposed to 10 µM LGK-974 (a Wnt3a inhibitor) for 48 hours. The viability of cells was determined using MTT method. The ability to differentiation into endothelial cells was assessed by measuring expression and protein levels of VE-Cadherin and vWF using real-time PCR and western blotting. Results: The inhibition of Wnt3a in H9C2 cells could significantly reduce cell survival rate after 48 hours compared to the control cells (p<0.05). Based on data, expression and protein levels of VE-Cadherin and vWF were significantly diminished in group incubated with LGK-974 Conclusion: The inhibition of Wnt3a can suppress the angiogenic potential of rat cardio myoblasts

Isolation and Differentiation of Neural Stem/Progenitor Cells From Subventricular Zone of One Adult Rat

Introduction: In adult mammalian brain, neural stem cells are isolated from both the dentate gyrus and subventricular zone. This study aimed to isolate neural stem cells from adult rat subventricular zone and differentiate them into neurons and astrocytes. Methods: In this study, the whole brain was removed after full anesthesia and creating cervical dislocation. Under a microscope, subv...

Human Trophoblast Progenitor Cells Express and Release Angiogenic Factors

Trophoblast stem cells develop from polar trophoectoderm and give rise to the cells that generate the placenta. Trophoblast cells are responsible for the uterinal invasion and vascular remodeling during the implantation of the embryo. However this knowledge is not yet to be confirmed for trophoblast progenitor cells (TPCs). In this study, we aimed to demonstrate that human TPCs (hTPCs) express ...

Osteogenic Differentiation Capacity of Human Skeletal Muscle-Derived Progenitor Cells

Heterotopic ossification (HO) is defined as the formation of ectopic bone in soft tissue outside the skeletal tissue. HO is thought to result from aberrant differentiation of osteogenic progenitors within skeletal muscle. However, the precise origin of HO is still unclear. Skeletal muscle contains two kinds of progenitor cells, myogenic progenitors and mesenchymal progenitors. Myogenic and mese...

Response of Endothelial Progenitor Cells and Expression of Angiogenic Cytokine Genes in Preconditioning with Resistance Training in Elderly Men

Introduction: Impairment in the endothelial function starts with aging; however, it can be reduced or prevented by regular exercise. This study aimed to investigate the response and compatibility of endothelial progenitor cells and to evaluate the gene expression of hypoxia-inducible factor-1 (HIF-1) and vascular endothelial growth factor (VEGF) after eight weeks of resistance training in the e...

Activation and Inhibition of The Wnt3A Signaling Pathway in Buffalo (Bubalus bubalis) Embryonic Stem Cells: Effects of WNT3A, Bio and Dkk1

Background This research studies the effects of activation and inhibition of Wnt3A signaling pathway in buffalo (Bubalus bubalis) embryonic stem (ES) cell-like cells. MaterialsAndMethods To carry on this experimental study, the effects of activation and inhibition of Wnt3A signaling in buffalo ES cell-like cells were examined using Bio (0.5 mM) combined with WNT3A (200 ng/ml), as an activator, ...

Smad1 transcription factor integrates BMP2 and Wnt3a signals in migrating cardiac progenitor cells.

In vertebrate embryos, cardiac progenitor cells (CPCs) undergo long-range migration after emerging from the primitive streak during gastrulation. Together with other mesoderm progenitors, they migrate laterally and then toward the ventral midline, where they form the heart. Signals controlling the migration of different progenitor cell populations during gastrulation are poorly understood. Seve...
